Genomically Recoded and Optimized Streptomyces Strains

Description of the project offered: 
Members of genus Streptomyces are one of the most prolific producers of natural products and enzymes of enormous practical value for human well-being. They also serve as a simple prokaryotic model to understand complex processes of morphological development. The production of bioactive compounds and life cycle progression in streptomycetes are linked and coordinated by a complex network of regulators acting at all levels of gene expression. There is strong interest in industry and systems biology to develop engineered actinomycetes that are simpler than the wild type ones in some aspects (for example, smaller genomes, rewiring of regulatory pathways). Here we propose to generate a set of streptomycetes with simplified genetic code and tuned translational accuracy. These strains will be built upon genomes of Streptomyces albus J1074 and S. violaceusniger strain SPC6, which are convenient exression hosts because of naturally minimal genomes and fast growth. In GC-rich streptomycete genomes leucyl codon TTA is extremely rare, and present only in genes for auxiliary functions (for example, in 6.8 Mb genome of J1074 there are only 75 TTA codons) We will delete all TTA-containing genes which did not impose fitness costs on the strains; in "fitness-costly" genes the TTA codons will be substituted with a common synonym. The generated strains will further be manipulated to introduce mutations into gene rpsL for ribosomal protein S12, known to exert pleiotropic effects on Streptomyces biology. Our work will provide first example of genomically recoded actinobacterium, helping answer a number questions of both basic (roles of synonymous codons usage in evolution; genetic code expansion; regulagtory functions of the ribosome) and practical nature (generation of novel chassis for biomolecula production; development of UUA-tRNA -based systems for introduction of unnatural aminoacids)
