Bacterial Production of Carbon Nanotube

Description of the project offered: 
The basic idea is the creation of a mutant bacterium of Shewanella strain that is able to assemble carbon nanotubes from organic precursors.

The idea of the research project comes from some studies conducted on different strains of the bacterium Shewanella. In these studies (carried out by different laboratories) are shown three special abilities that these bacterial strains possess:
1. Shewanella putrefaciens and oneidensis can assemble arsenic sulfide nanotubes when they are in the presence of arsenic and sulfur, as a mechanism of protection against the toxic effects of these elements.
2. The same bacterial strains have been found in areas with high presence of hydrocarbons (some samples were taken during extraction wells spills ) It was found in fact that these organisms have the ability to use hydrocarbons such as electron donors in anaerobic respiration (in redox reactions).
3. In the last years some laboratories produced graphene sheets (the size of a few micrometers) using some strains of Shewanella (through redox chemistry) from graphene oxide.

To obtain this result it is advisabile to follow two parallel lines of research.

1. Starting coltures of Shewanella strains without the ability of assembling As-S nanotubes ( with plasmids isolated from original strain and randomly mutated) in medium with organic toxic substances in sub-lethal levels to induce the mechanism assembly of carbon nanotubes .

2. Analysis of the genome ( and Rna amplification), isolation of polysaccharide complex sequence that assemble arsenic sulfide nanotubes in the original strain, in order to explain the assembling process; wiht the obtained sequence assembling of random mutant plasmids.

Description of the collaboration sought: 
As private person, for the project I want to achieve, I need a host structure (university or research center) with the focus on biotechnology applied to bacteria. For this segment of research (basic research), it is preferable a university or state organization that has the possibility of using a network of laboratories of biology and chemistry; this way it can be supported all phases of the project in one main lab and then use external laboratories or resources for specific tests (sequencing, protein complex isolation and analysis of the chemical structure of the nanotubes ). The criteria for choosing the right center will be: the ability of the working group (resident or in call ), the technical capabilities (equipment and instrument) and the possibility of technology transfer.
